You should be able to do this. We've built slave nodes in VirtualBox as part of our Pools of Virtual Boxes project with no major problems.
If you go with VirtualBox for testing, I recommend creating internal network interfaces and statically assigning IP addresses and host names for each virtual machine. Make sure all the host names you use are in /etc/hosts on each machine. Aside from that, installing a Linux distribution and Condor in the virtual machines is straightforward.
